Monitoring of trace metal contamination in the Souss estuary (South Morocco) using the clams Cerastoderma edule and Scrobicularia plana ArchiMer
Anajjar, E; Chiffoleau, Jean-francois; Bergayou, H; Moukrim, A; Burgeot, Thierry; Cheggour, M.
Concentrations of seven metals (Cd, Cu, Cr, Pb, Ni, Zn, Hg) were monitored monthly during 2 years in two bivalves, Cerastoderma edule and Scrobicularia plana collected from the Souss estuary (South Morocco) before and after the commissioning of a sewage treatment plant in November 2002. According to trace metals, changes in concentrations were observed mainly in S. plana, characterised by a decrease of Cu, Hg and Pb when Cd was clearly increasing. Conversely, no clear changes were observed in C. edule during the same periods. This study suggests the use of S. plana in the monitoring of trace metal contamination of such coastal marine environments.

In the scope of the study of the "on-bottom/off-bottom" mortality model in the south of the Marennes Oléron Bay, the result of the 2003 multidisciplinary study (Dynamor) appears to be the hypothesis of an acute stress, "ultimate" trigger of the mortality episodes in June. In 2004 we aim to look for the nature of this stress through the physico-chemical analysis of the sediment (Eh, pH, sulphides and ammoniacal nitrogen), the organic matter and chlorophyll load on the surface of the sediment between April and July. Herbicides are also measured in the water column, south of the bay (Seudre), during the same period. A "precocious" (around the first week of June) and "weak" (lower mortality rate on this working site since 1997) mortality peak happens when the...

Multigenerational exposure of the microalga Tetraselmis suecica to diuron leads to spontaneous long-term strain adaptation. ArchiMer
Stachowski-haberkorn, Sabine; Jerome, Marc; Rouxel, Julien; Khelifi, Celia; Rince, Maeva; Burgeot, Thierry.
To investigate the ability of microalgae to develop stable, long-term resistance to herbicides, the marine microalga Tetraselmis suecica was exposed to the herbicide diuron (5 µg/L) for a 43-generation exposure period followed by a 12-generation depuration phase. During the first 25 generations, diuron-exposed cultures showed doubling times ranging from 1.95 to 2.6 days, which was 2 to 2.5-fold longer than control cultures. Between generations 25 and 38, during diuron exposure, two out of the three exposed cultures exhibited a spontaneous drop in doubling time. These results provided evidence of culture adaptation to diuron. To assess persistence of the diuron adaptation observed on growth performance, one of the adapted cultures (D3) was maintained for 12...

Nutrition and reproduction are key parameters in the interaction process leading to Crassostrea gigas oyster summer mortality in France ArchiMer

Complex interactions between oyster, environment and pathogens were observed during the MOREST (2001-2005) project on summer mortality events of Crassostrea gigas oysters in France. The talk will show how nutrition level increasing reproductive intensity generates a risk for pathogen infection under synergistic effect of temperature and stress. Moreover, a genetic component is associated to specific reproductive performances and spawning behaviour. To better understand origin of such resistance or susceptibility to summer mortality, other characteristics as stress susceptibility, energy management and hemocyte activities of oysters from the two phenotypes were compared. This first comparison suggests interactions between stress, reproduction and defence...

Pacific oyster (Crassostrea gigas) hemocyte are not affected by a mixture of pesticides in short-term in vitro assays ArchiMer
Moreau, Pierrick; Burgeot, Thierry; Renault, Tristan.
Pesticides are frequently detected in estuaries among the pollutants found in estuarine and coastal areas and may have major ecological consequences. They could endanger organism growth, reproduction or survival. In the context of high mortality outbreaks affecting Pacific oysters, Crassostrea gigas, in France since 2008, it appears of importance to determine the putative effects of pesticides on oyster susceptibility to infectious agents. Massive mortality outbreaks reported in this species, mainly in spring and summer, may suggest an important role played by the seasonal use of pesticides and freshwater input in estuarine areas where oyster farms are frequently located. To understand the impact of some pesticides detected in French waters, their effects...

PAH metabolites in fish bile: from the Seine Estuary to Iceland ArchiMer
Kammann, U.; Akcha, Farida; Budzinski, H.; Burgeot, Thierry; Gubbins, M. J.; Lang, T.; Le Menach, K.; Vethaak, A. D.; Hylland, K..
Polycyclic aromatic hydrocarbons (PAH) are environmental contaminants that pose significant risk to health of fish. The International Workshop on Integrated Assessment of Contaminant Impacts on the North Sea (ICON) provided the framework to investigate biomarker responses as well as contaminant concentrations side by side in marine ecosystems. Concentrations of the main PAH metabolites 1-hydroxypyrene, 1-hydroxyphenanthren and 3-hydroxybenzo(a)pyrene were determined in bile by HPLC with fluorescence detection. Fish species under investigation were dab (Limanda limanda), flounder (Platichthys flesus) and haddock (Melanogrammus aeglefinus). A contamination gradient was demonstrated from the low contaminated waters of Iceland and off-shore regions of the...

Parental exposure to environmental concentrations of diuron leads to aneuploidy in embryos of the Pacific oyster, as evidenced by fluorescent in situ hybridization ArchiMer
Barranger, Audrey; Benabdelmouna, Abdellah; Degremont, Lionel; Burgeot, Thierry; Akcha, Farida.
Changes in normal chromosome numbers (i.e. aneuploidy) due to abnormal chromosome segregation may arise either spontaneously or as a result of chemical/radiation exposure, particularly during cell division. Coastal ecosystems are continuously subjected to various contaminants originating from urban, industrial and agricultural activities. Genotoxicity is common to several families of major environmental pollutants, including pesticides, which therefore represent a potential important environmental hazard for marine organisms. A previous study demonstrated the vertical transmission of DNA damage by subjecting oyster genitors to short-term exposure to the herbicide diuron at environmental concentrations during gametogenesis. In this paper, Fluorescent in...

Parental exposure to the herbicide diuron results in oxidative DNA damage to germinal cells of the Pacific oyster Crassostrea gigas ArchiMer
Barranger, Audrey; Heude-berthelin, Clothilde; Rouxel, Julien; Adeline, Beatrice; Benabdelmouna, Abdellah; Burgeot, Thierry; Akcha, Farida.
Chemical pollution by pesticides has been identified as a possible contributing factor to the massive mortality outbreaks observed in Crassostrea gigas for several years. A previous study demonstrated the vertical transmission of DNA damage by subjecting oyster genitors to the herbicide diuron at environmental concentrations during gametogenesis. This trans-generational effect occurs through damage to genitor-exposed gametes, as measured by the comet-assay. The presence of DNA damage in gametes could be linked to the formation of DNA damage in other germ cells. In order to explore this question, the levels and cell distribution of the oxidized base lesion 8-oxodGuo were studied in the gonads of exposed genitors. High-performance liquid chromatography...

Pesticides and Ostreid Herpesvirus 1 Infection in the Pacific Oyster, Crassostrea gigas ArchiMer
Moreau, Pierrick; Faury, Nicole; Burgeot, Thierry; Renault, Tristan.
Since 2008, mass mortality outbreaks have been reported in all French regions producing Pacific oysters, and in several Member States of the European Union. These mass mortality events of Pacific oysters are related to OsHV-1 infection. They occur during spring and summer periods leaving suspect the quality of the marine environment and the role of seasonal use of pesticides associated with the arrival of freshwater in oyster rearing areas. Pesticides have been also detected in French coastal waters, especially in areas of oyster production. Using PMA real-time PCR we showed that a mixture of 14 pesticides has no effect on the integrity of virus capsids from viral suspension in the conditions tested. A contact of oysters with this pesticide mixture was...

Pollutant effects on Pacific oyster, Crassostrea gigas (Thunberg), hemocytes: Screening of 23 molecules using flow cytometry ArchiMer
Gagnaire, Beatrice; Thomas Guyon, H; Burgeot, Thierry; Renault, Tristan.
The shellfish industry is an important economic activity in France, occurring mostly in estuarine zones subject to pollution due to anthropogenic activities. The harmful effects of pollutants on species inhabiting these estuarine zones are not well known. Among marine species, bivalve mollusks---particularly Pacific oyster, Crassostrea gigas-may serve a model of interest. The species is sedentary and filter-feeding, which favors bioaccumulation of pollutants in their tissues. Oysters may be suitable for studies on disturbance by pollutants of physiological activities, among which defense mechanisms are poorly documented in bivalves. In this study, effects of pollutants on hemocyte functions were monitored in Pacific oyster, C. gigas. Hemocytes were exposed...

Polycylic aromatic hydrocarbons (PAHs) are ubiquitous contaminants in the marine environment Their toxicity is mainly linked to the ability of marine species to biotransform them into reactive metabolites PAHs are thus often detected at trace levels in animal tissues For biomonitoring purposes this findings have two main consequences (i) the determination of the PAH tissue concentration is not suitable for the evaluation of individual exposure to PAHs (ii) It can explain sometimes the lack of correlations obtained with relevant markers of toxicity such as genotoxicity biomarkers The aim of the present study was to better investigate the link between PAH exposure and genotoxicity in marine flatfish During a laboratory experiment juvenile soles were exposed...

Human‐mediated transport creates secondary contacts between genetically differentiated lineages, bringing new opportunities for gene exchange. When similar introductions occur in different places, they provide informally replicated experiments for studying hybridisation. We here examined 4,279 Mytilus mussels, sampled in Europe and genotyped with 77 ancestry‐informative markers. We identified a type of introduced mussels, called “dock mussels,” associated with port habitats and displaying a particular genetic signal of admixture between M. edulis and the Mediterranean lineage of M. galloprovincialis. These mussels exhibit similarities in their ancestry compositions, regardless of the local native genetic backgrounds and the distance separating colonised...

Sampling design optimization for EROD measurements in fish ArchiMer
Beliaeff, Benoit; Burgeot, Thierry.
Activity of 7-ethoxyresorufin-O-deethylase (EROD) in fish is certainly the best-studied biomarker of exposure applied in the field to evaluate biological effects of contamination in the marine environment. Since 1991, a feasibility study for a monitoring network using this biomarker of exposure has been conducted along French coasts. Using data obtained during several cruises, this study aims to determine the number of fish required to detect a given difference between 2 mean EROD activities, i.e. to achieve an a priori fixed statistical power (l-beta) given significance level (alpha), variance estimations and projected ratio of unequal sample sizes (k). Mean EROD activity and standard error were estimated at each of 82 sampling stations. The...

Economic and social developments have taken place at the expense of the health of the environment, both locally and on a global scale. In an attempt to better understand the large-scale effects of pollution and other stressors like climate change on the health status of Mytilus edulis, mussels were collected during the first two weeks of June 2005 at three sites (one pristine and two affected by pollution) located in each of the regions of the Canadian West Coast, the St. Lawrence estuary, the Atlantic East Coast and the northwestern coast of France, covering a total distance of some I 1000 km. The mussels were analyzed for morphologic integrity (condition factor), gametogenic activity (gonado-somatic and gonad maturation index, vitellogenin(Vtg)-like...

Study of genetic damage in the japanese oyster induced by an environmentally-relevant exposure to diuron: evidence of vertical transmission of dna damage ArchiMer
Barranger, Audrey; Akcha, Farida; Rouxel, Julien; Brizard, Raphael; Maurouard, Elise; Pallud, Marie; Menard, Dominique; Tapie, N.; Budzinski, Helene; Burgeot, Thierry; Benabdelmouna, Abdellah.
Pesticides represent a major proportion of the chemical pollutants detected in French coastal waters and hence a significant environmental risk with regards to marine organisms. Commercially-raised bivalves are particularly exposed to pollutants, among them pesticides, as shellfish farming zones are subject to considerable pressure from agricultural activities on the mainland. The aims of this study were to determine (1) the genotoxic effects of diuron exposure on oyster genitors and (2) the possible transmission of damaged DNA to offspring and its repercussions on oyster fitness. To investigate these points, oysters were exposed to concentrations of diuron close to those detected in the Marennes-Oleron Basin (two 7-day exposure pulses at 0.4 and 0.6 μg...

An oceanographic cruise in 1991 along the English Channel and Atlantic coasts of France indicated the value of dragonet Callionymus lyra Linnaeus, 1758 as a target species for monitoring biological effects. Ethoxyresorufin-O-deethylase measurements in this species provided a first assessment of the effects of hydrocarbons (PCB, PAH, dioxins) on the marine environment. The specific behavior of dragonet was defined by biochemical characterization. Biological data were obtained during 3 additional oceanographic cruises to study changes in the abundance and distribution of the species in the English Channel and the Bay of Biscay. This first assessment, carried out in the context of the National Observation Network, indicated that pollutants do not have...

The integrated biomarker response revisited: optimization to avoid misuse ArchiMer
Devin, S.; Burgeot, Thierry; Giamberini, L.; Minguez, L.; Pain-devin, S..
The growing need to evaluate the quality of aquatic ecosystems led to the development of numerous monitoring tools. Among them, the development of biomarker-based procedures, that combine precocity and relevance, is recommended. However, multi-biomarker approaches are often hard to interpret, and produce results that are not easy to integrate in the environmental policies framework. Integrative index have been developed, and one of the most used is the integrated biomarker response (IBR). However, an analysis of available literature demonstrated that the IBR suffers from a frequent misuse and a bias in its calculation. Then, we propose here a new calculation method based on both a more simple formula and a permutation procedure. Together, these...
